新宿区歌舞伎町の路地にひっそりと建つ『聖まごころ病院』。その土地柄、救急外来を訪れる患者の多くは、貧しい路上生活者や在留外国人、ホストや風俗嬢など、一般の病院と違い、その特殊な“ワケあり”背景を持った患者たちが集う。そこに、突如としてアメリカ国籍の元軍医である謎の女性が歌舞伎町に降臨する。彼女は、軍医経験を持つ女医のヨウコ・ニシ・フリーマン。やがて彼女は、歌舞伎町で繰り広げられるさまざまな闇を明るく照らし出す存在となっていく―。そして、年老いた叔父の病院に勤務する美容皮膚科医の高峰享（たかみね・とおる）がヨウコと出会い、歯車が動き始める―。時代とともに大きな変貌を遂げていく歌舞伎町の中で、ポツリと小さく存在し、取り残されたボロい病院。ネオンを光らせ、人々の欲がうごめく夜の街・歌舞伎町の中で生きる人たちに、小さな病院の光が、ひとつの心のよりどころとして、病院にいる個性豊かな医師たちとともに街に明かりをともしていく―。
